Dupuis was a King's Scholar and went on to King's in 1815. He was a fellow there from 1818 - 1831 but returned to Eton as an assistant master in 1819. In 1834 he became Lower Master but was elected a Fellow of Eton in 1838 and Vice-Provost in 1868. In addition he held the livings of Hemingby, Lincs, from 1831 - 1840, Creeting from 1840 - 1862, and Worplesdon from 1862 - 1878. His wife Julia was the daughter of William Roberts, Vice-Provost 1819 - 1833.
