Son of Sir Anthony Rous of Halton St Dominick, Cornwall, Rous was educated at Broadgate Hall, Oxford before graduating from the University of Leiden. He entered the Middle Temple in 1601. In 1625 - 1626 he was elected MP for Truro, which he also represented in the Short and Long Parliaments. In 1628 - 1629 he represented Tregony. He was nominated an assessor of the Westminster Assembly in 1643 and appointed Provost of Eton in February 1644. He held a number of Commonwealth posts and was elected Speaker of the Little Parliament of 1653, a member of Cromwell's Council of State, and in 1657, was created a lord of parliament. He wrote numerous theological works.
